Molecular Targeting of Human Papillomavirus oncogene expression using RNA interference Technology

چکیده
tissue is accomplished by removal of keratinocytes harboring HPV. These are the conventional approaches for treatment of cervical cancer. Prophylactic vaccines are available and should be administered prior to viral exposure. Initiatives for vaccination have been undertaken by the governments and also non-governmental agencies. Attempts are being made to develop therapeutic vaccines that would treat prevailing HPV infection. Use of immunomodulators like interferon and imiquimod induce cytokine production to increase host immune response against HPV infection [5]. Many antioxidants and poly herbal formulations are also in use. Molecular targeting of HPV gene expression using RNA interference technology includes use of antisense oligonucleotides, ribozymes, short interfering RNAs (siRNA), short hairpin RNA (shRNA) etc. The present review is a criMolecular Targeting of Human Papillomavirus oncogene expression using RNA interference Technology Sarmistha Sen Raychaudhuri 1,* and Sreejata Raychaudhuri 2,†

RNA interference against human papillomavirus oncogenes in cervical cancer cells results in increased sensitivity to cisplatin.
Targeted inhibition of oncogenes in tumor cells is a rational approach toward the development of cancer therapies based on RNA interference (RNAi). Tumors caused by human papillomavirus (HPV) infection are an ideal model system for RNAi-based cancer therapies because the oncogenes that cause cervical cancer, E6 and E7, are expressed only in cancerous cells. We investigated whether targeting HPV...
متن کاملTargeting the human papillomavirus E6 and E7 oncogenes through expression of the bovine papillomavirus type 1 E2 protein stimulates cellular motility.
Expression of the high-risk human papillomavirus (HPV) E6 and E7 oncogenes is essential for the initiation and maintenance of cervical cancer. The repression of both was previously shown to result in activation of their respective tumor suppressor targets, p53 and pRb, and subsequent senescence induction in cervical cancer cells. Consequently, viral oncogene suppression is a promising approach ...
متن کاملSuppressive Effect of Constructed shRNAs against Apollon Induces Apoptosis and Growth Inhibition in the HeLa Cell Line
Background: Cervical cancer is the second most common female cancer worldwide. Inhibitors of apoptosis proteins (IAPs) block apoptosis; therefore, therapeutic strategies targeting IAPs have attracted the interest of researchers in recent years. Apollon, a member of IAPs, inhibits apoptosis and cell death. RNA interference is a pathway in which small interfering RNA (siRNA) or shRNA (short hairp...
متن کاملThe E7 oncoprotein is translated from spliced E6*I transcripts in high-risk human papillomavirus type 16- or type 18-positive cervical cancer cell lines via translation reinitiation.
High-risk human papillomaviruses (HPVs) encode two viral oncoproteins, E6 and E7, from a single bicistronic pre-mRNA containing three exons and two introns. Retention of intron 1 in the E6 coding region is essential for production of the full-length E6 oncoprotein. However, splicing of intron 1 is extremely efficient in cervical cancer cells, leading to the production of a spliced transcript, E...
متن کاملOncogenic Human Papillomaviruses Activate the Tumor-Associated Lens Epithelial-Derived Growth Factor (LEDGF) Gene
The expression of the human papillomavirus (HPV) E6/E7 oncogenes is crucial for HPV-induced malignant cell transformation. The identification of cellular targets attacked by the HPV oncogenes is critical for our understanding of the molecular mechanisms of HPV-associated carcinogenesis and may open novel therapeutic opportunities. Here, we identify the Lens Epithelial-Derived Growth Factor (LED...
